# Southern Blot
"Dot Blot -> Southern Blot" is the DNA equivalent of "ELISA -> Western Blot"
- **Reagents:**
- Agarose Gel
- Probes
- Restriction Enzymes
Probes are useful for locating SNPs and STRs which are both highly variable changes in genomes that can be associated with diseases:
Both can be used for:
- Pedigree/Inheritance Analysis
- Disease Diagnosis
- Genetic Markers when Mapping Genes
- Paternity Testing
**Restriction Fragment Length Polymorphisms (RFLP):** Involves analyzing *single nucleotide polymorphisms (SNPs)*, which change the restriction enzyme digestion site and thus pattern on a Southern blot.
**Variable Number of Tandem Repeats (VNTR):** Involves analyzing *short tandem repeats (STRs)*, which are highly variable in the population.
- The amount of repeats can *vary between the two alleles* within a single individual
